ras経路による分裂酵母細胞の分化制御

研究概要	分裂酵母のRasタンパク質が活性化することにより、接合フェロモン受容の経路上にあるMAPKカスケードの反応性が上昇し、シグナルが伝わりやすくなることが知られていた。一方、Rasを欠く細胞は細胞形態が丸くなること、そしてこの形態変化は上述のMAPKカスケードとは無関係であることから、分裂酵母Rasには第二の標的が存在するのではないかと考えられたきた。本研究における一つの焦点として、我々が先に単離していた、ras欠損変異株と細胞形態および接合不能性の表現型においてよく類似するrallおよびral3突然変異株に注目し、rallおよびral3遺伝子の構造決定を行った。その結果、rall遺伝子は出芽酵母のCDC24遺伝子と相同性をもち、低分子量GTP結合タンパク質のGDP-GTP交換因子と考えられる構造をしていた。一方ral3遺伝子は出芽酵母のBEM1遺伝子と相同性をもっていた。BEM1遺伝子は芽のでる部位の決定に関わっており、CDC24ともども細胞の極性決定に関与すると考えられている。出芽酵母でこれらの因子は、rasとよく似た構造をもつ動物遺伝子Krev-1の出芽酵母におけるホモログであるRSR1と相互作用することが知られているが、上述の結果は、RSR1に相当するものが見つからない分裂酵母では、Rasタンパク質がこれらの因子と相互作用して細胞の極性決定に関わっている可能性を強く示唆した。この研究成果はスペインでの国際会議で発表したが、同様の研究結果がM.Wiglerのグループによっても得られている。本研究ではまた、分裂酵母でRasタンパク質のGDP-GTP交換因子をコードするste6遺伝子の発現が、接合フェロモン受容シグナルにより増強されることを明らかにした。この結果は、Rasの活性化を介して、接合フェロモン受容のシグナルがポジティブフィードバックにより自己増強することを示している。
