研究概要	生体内では、多種類の細胞が精緻な三次元構造を形成し、細胞間相互作用のもとに増殖と分化のホメオスタシスが維持されている。本研究はそのような組織構築の分子メカニズムを、発がんの最大の標的組織である上皮組織について分析しようとするものである。本年度は以下の研究業績を得た。一)三次元培養系の樹立: 組織構築のモデルとして線維芽細胞とケラチノサイトの再構成による三次元培養系を確立し、細胞間相互作用を検討した。線維芽細胞を低張処理によって殺したコラーゲンゲル上では、ケラチノサイトの増殖が著しく抑えられた。この時の“paracrine"増殖因子はHGFではなかった。さらに、胎生期マウス皮膚を器官培養し形態形成、分化発現を試験管内で再現する実験系を樹立した。増殖因子の毛芽発生に及ぼす影響を検討した。 2)ゲラチノサイト分化のシグナル伝達経路の分析: マウス皮膚cDNAライブラリーから分離したCキナーゼη分子種は、上皮組織に豊富に存在し上皮細胞の分化に伴って発現する。バキュロウイルス法によって得たη分子種は、コレステロール硫酸によって活性化されることが分った。コレステロール硫酸は、ケラチノサイトの分化に伴って作られるところから、この物質はη分子種のセカンドメッセンジャーとして働いているものと思われる。コレステロール硫酸は、ケラチノサイトおよびそのがん化細胞の増殖を抑え分化を誘導する。また、マウス皮膚二段階発がん実験系では発がんプロモーションを抑制することが明らかになった。以上の結果、ケラチノサイトの分化シグナル伝達に際して、コレステロール硫酸→Cキナーゼη分子種の経路が中心を占めていることが明らかになった。
