ジャクスタクリン機構を介した細胞膜結合型細胞増殖因子と癌細胞増殖

研究概要

本年度は、膜結合型HB-EGF(proHB-EGF)のジャクスタクリン活性の発現機構を中心にproHB-EGFの膜タンパク質としての機能を解析し、さらに癌組織でのproHB-EGFの発現を検討し、以下の結果を得た。
1.膜結合型へパリン結合性EGF様増殖因(proHB-EGF)がジャクスタクリン増殖因子としての活性を発現するためには別の膜蛋白質CD9と複合体を形成することが必須であることを明らかにした。
2.proHB-EGFは、ジャクスタクリン増殖因子としてだけでなくジフテリア毒素受容体としても機能することを明らかにした。
3.proHB-EGFから分泌型HB-EGFへの変換はC-キナーゼを介する経路で制御されており、ホルボールエステル等によるC-キナーゼの活性化がproHB-EGFのジャクスタクリン因子からパラクリン因子への変換のスイッチであることを明らかにした。
4.ヒト癌組織でのproHB-EGFの発現と産生をノーザンブロット解析と免疫組織染色により討した結果、ヒト正常肝組織でのproHB-EGFの産生は極めて低いレベルであるのに対し、癌組織でのproHB-EGFの産生がきわめて高くなっていることを明らかにした。
