細胞接着因子様の細胞外ドメインを持つ新規レセプター型チロシキナーゼの機能

研究概要	1994年、私達は脳に特異的に高発現する新規なレセプター型チロシンキナーゼ、"Sky",をヒトcDNAライブラリーよりクローニングした。Skyは細胞内ドメインにチロシンキナーゼ構造をもち、細胞外ドメインにイムノグロブリン様ドメインとフィブロネクチンタイプIIIドメインをもつ。これらの細胞外ドメイン構造は神経細胞接着因子(NCAMs)やプロテインチロシンフォスファターゼなどの細胞接着因子に共通にみられる構造で、Skyは神経系における細胞接着に関与する可能性が考えられる。本研究では、Skyの機能を明らかにするため、まずラットSkyのcDNAをクローニングし、これをプローブとして、in situ hybridizationによりSky mRNAの脳内発現分布を明らかにした。この結果、skyは海馬CA1領域、小脳顆粒細胞層、嗅球内顆粒層、帯状回、紐蓋、扁桃体、大脳皮質など神経細胞の密に存在する部位での高い発現が観測された。またSkyに対する特異抗体を用いて、Skyの自己リン酸化活性化及びSrcファミリーキナーゼとの会合について明らかにした。さらにSkyの機能を解明するためSkyのリガンドを検出する系を作成した。すなわち、Skyの細胞外ドメインとヒトIgGのFc領域の融合蛋白質を発現させ、これを用いた発現クローニング及び、アフィニティークロマトグラフィーによるリガンドの精製を現在進めている。
