ラットグルタチオントランスフェラーゼP遺伝子の肝癌特異的発現機構

研究概要	われわれは、GPEIに結合する因子の解析を行い、肝癌細胞AH66と正常肝で比較検討したところ複数の因子がDPEIに結合することを見いだした。フットプリント法の結果より、1)GPEIのTRE-like配列に結合する因子、2)GPEIのTRE-like配列の20-50bp上流に結合する複数の因子を同定した。1)の因子は肝癌と正常肝でフットプリント上では差が認められないが、2)の因子は両者の間でフットプリント上で明らかな差が認められる。1)の因子はさらにDEAE-Sepharoseに結合するもの(C1)とそうでないもの(C2)に分かれる。これらの結果、GPEIに結合する因子は少なくとも4種類はあることが明らかとなった。 1)の因子(C1及びC2)をDEAE-Sepharoseによって分画後、さらに二つのカラムで部分精製し、それらのDNA結合能をゲルシフト法及びフットプリント法で検討した。また、それぞれの分画をc-jun及びmafの抗体を用いたimmunoblotにて検討した。フットプリント法を用いた結合実験で、C1、C2共にGST-Pの転写に重要なcore GPEIの領域に結合することが分かった。又、ゲルシフト法及びフットプリント法で競合実験を行った結果、両因子共にcore GPEIのG→Tの点突然変異によって結合が著しく低下し、GPEI変異体の転写活性とC1、C2に対する結合活性とは特異性が一致することが分かった。さらにimmunoblotの結果、C1にはc-jun、maf共に検出されなかったが、C2にはc-junが検出された。また、C2にはmafの抗体と反応する55kDaのタンパク質が検出され、分子量より、新しいmafファミリーのタンパク質と考えられる。部分分画した再構成in vitro転写系で両因子の転写活性の検出を試みたが、GTS-Pプロモーターの活性が低いため未だ明確な結果は得られていない。以上より、C1、C2共にGST-Pの転写活性化に関与している可能性が大きく、それぞれの因子をさらに精製し、それらのcDNAを単離する予定である。
