研究概要

Ras前がん遺伝子産物は細胞の増殖・分化における様々な局面において重要な役割を果たしている。RasのGTPアーゼ活性を促進することによりRasシグナル伝達系を負に制御する因子として、従来GAPおよびNF1の二種の因子が知られていたが、我々はGAPおよびNF1とは分子量および抗原性をことにする新しいGTPアーゼ活性促進因子の精製に成功した。さらに部分アミノ酸配列を基に新規GTPアーゼ活性促進因子の遺伝子を単離し、その全構造を決定した。
新規因子はその分子内部にRasのGTPアーゼ活性を促進する領域の他にリン脂質結合部位およびPHドメインを有しており、種々のシグナル伝達系によって制御され、かつシグナル伝達系間のクロストークに関与する可能性を示唆した。
Rasシグナル伝達系の制御機構を更に検討する過程で、新たなRas活性化因子の遺伝子クローニングを行ない、この因子が二つのアダプター因子に結合すること、酵母菌の中でRas活性化能を有することを示した。またHL-60細胞がマクロファージ様の細胞に分化する際にRasが活性化されること、Ras変異体がHL-60細胞の分化を抑制することから、HL-60細胞の分化過程にRasが必須であることを示した。
