研究概要

本年度の各班員の成果は以下個別に列挙するとおりである。
1.アデノウイルス癌遺伝子を導入して癌化させた細胞に、特異的にアポトーシスを起こす物質としてleptofuranin群化合物を単離した。この化合物は、癌細胞の細胞死を特異的に誘起したが、正常細胞に対しては増殖を抑制したが、毒性は示さなかった。
2.海洋らん藻から単離された細胞分裂阻害剤curacin Aの部分構造および類縁体10種を合成し、チューブリンへの作用を調べた結果、合成したcuracin A類縁体のチューブリン重合阻害活性はIC_<50>値2.5μMであったが、ヘテロ環部分および脂肪鎖部分のいずれも、それのみでは活性を示さなかった。
3.イトマキヒトデの受精を阻害しないで、胚発生を嚢胚形成期で選択的に阻害する活性物質を探索した結果、Hippospongia属海綿から新規トリテルペンを得た。本化合物はヒト白血病細胞MOLT-4の増殖をLD_<50>値50nM、ヒトミエロイドK-562細胞の増殖をLD_<50>値40nMと強く阻害した。
4.Aspergillus fumigatusの培養液から、新規ジケトピペラジン化合物スピロトリプロスタチンA、BとサイクロトリプロスタチンA-Dを見出した。これらの化合物は、マウス乳癌細胞FM3Aの温度感受性変異株tsFT210細胞の細胞周期をM期で停止させた。サイクロトリプロスタチンAは、in vitroでチューブリンの重合を阻害した。
5.熱帯産植物の果実のメタノール抽出物中にバフンウニの受精卵に対する第一卵割阻害活性を認め、活性化合物としてL-(-)-di-n-buty1 malateを16mgを得た(MIC:8μg/ml)。誘導体を各種合成し活性の違いを検討した結果、L-(-)-di-n-penty1 malateが最も活性が強いことが判明した。
