研究概要

1.造血器腫瘍における転座関連遺伝子6個を新たに単離/同定した。
2.転座関連遺伝子の構造/機能解析
a)AML1遺伝子のゲノム構造を決定した。b)15,17転座により形成されるPML-RAR蛋白質はPML蛋白質と複合体を形成し、ドミナント・ネガティブ作用により白血病発症に寄与することが解った。c)MLL遺伝子構造を明らかにするとともに、この遺伝子と融合する他の遺伝子(LTG4,LTG9,LTG19)との構造比較を行った。d)BCL2トランスジェニックマウス実験より、14;18転座腫瘍の発症にはBCL2遺伝子を含んだ複数遺伝子の活性化が必要であることを証明した。e)p210^<bcr-ab1>トランスジェニックマウス系を樹立し、BCR-ABL遺伝子が腫瘍原性であることを初めて証明した。
3.転座型白血病/リンパ腫症の分子病態研究
a)22例の1;19転座白血病の検討によりE2A+PBX1融合遺伝子形成とp53遺伝子変異を持つ症例は予後が悪いことを証明した。b)び慢性リンパ腫に関与するBCL6再構成は74例の濾胞性リンパ腫の検討により本症の進展にも強く関連していることが示唆された。c)FISH法を用いた検討により二次性白血病/悪性リンパ腫には特定部位の遺伝子増幅が発症・進展に関与していることが示唆された。
4.間期核上で転座白血病が診断可能なFISH法の開発
a)血液塗沫標本上で8;21転座、9;22転座、11q23転座および14q32転座の診断が48時間以内に可能になった。b)治療後の残存白血病細胞の10^<-3>レベルで顕微鏡下に検出可能になった。
5.抗体による転座型白血病/リンパ腫症の診断
PRAD1,PML,RARA遺伝子産物に対する抗体作製に成功し,MCL,APLにおける抗体による診断が可能になった。
