酵素分子設計による抗ガン剤副作用軽減法の開発

研究概要

活性酸素が癌化学療法に重要な役割を担うが、同時にこれが心筋障害や腎障害のごとく、正常組織に対する副作用となり、抗がん剤の使用量を制限する主要因子となっている。したがって、癌化学療法では、その殺腫瘍細胞作用を減弱することなく副作用を如何に阻止軽減するかが重要な課題となっている。我々は、活性酸素毒性を個体レベルで組織細胞特異的に制御する活性酸素代謝酵素誘導体を開発し、分子から個体のレベルで実験を行い、以下の研究実績を得た。
1)遺伝子組み換えにより、ヒト細胞質型Superoxide Dismutase(SOD)を大量に発現する系を確立し、長時間血中を循環する病巣指向性SOD誘導体を開発した。さらに、遺伝子組み換えにより、SODのC-末側に塩基性アミノ酸に富むヘパリン結合性ハイブリッドSOD(HB-SOD)を発現させ、本酵素が血管内皮細胞に個体レベルで特異的に結合分布し、スーパーオキシドラジカルを有効に消去することを明らかにした。
2)上記のSOD誘導体に加え、積極的に糸球体濾過されて腎近位尿細管に特異的に集積する腎指向性SOD誘導体(AH-SOD)を開発した。
3)AH-SODがシスプラチン腎症における機能障害を阻止軽減することを確認した。
4)腹水肝癌細胞AH-130を腹腔内移植したラットにシスプラチンを投与し、その抗腫瘍作用と腎機能変化を解析した。解析の結果、シスプラチンがAH-SODの増殖を抑制すると同時に、その腎機能障害がAH-SODにより抑制されるうることが明らかになった。現在、その詳細な動力学的解析を行いつつある。
5)肝実質細胞やKupffer細胞をはじめとする正常の肝構成細胞はガラクトースやマンノースに対する特異的レセプターを有するが、腹水肝癌細胞や転移癌細胞はこれらのレセプターを持たない。今後、腹水肝癌細胞の肝転移モデル動物を作製し、抗ガン剤の肝内局所投与による抗腫瘍作用と正常肝組織細胞障害を比較検討すると同時に、肝細胞及び類洞細網内皮系細胞指向性SODによる正常細胞保護作用を解析する。
