がん遺伝子c-kit及びMLLを標的としたがん化学療法の基礎的研究

研究概要

がん遺伝子及びその産物は新しいがん治療法の標的となる可能性が高く、今後の化学療法にとって極めて有望な標的分子であり、そのための基礎的研究が必要である。
申請者らは、c-kit遺伝子(肺小細胞癌,乳癌)及びMLL遺伝子(乳児白血病,二次性白血病)をがん化学療法の標的分子モデルとして、将来的には、遺伝子レベルから蛋白レベルまでの種々の段階で腫瘍の増殖抑制・分化誘導に効果を示す薬剤等の検索を目指し、検討を行っている。本年度の成果は以下に要約される。
1.乳児白血病や二次性白血病の標的遺伝子であるMLL(mixed cell lineage leukemia)遺伝子の造腫瘍性に関して、MLL遺伝子の特定領域(Zinc finger領域のN端側)での分断が重要であることが、強く示唆される結果が得られた。
2.EμSV-プロモーターを持つ発現ベクターにて、MLL-LTG19及びMLL-LTG9トランスジェニックマウスを作出し1年以上経過観察中であるが、白血病の発癌は認めていない。
3.肺小細胞癌培養株のうちc-kit陽性株と陰性株を指標として、足場非依存性増殖能スクーリングシステムにて、c-kit陽性細胞に特異的増殖抑制効果を示す薬剤を見い出した。
緑膿菌外毒素結合ドメンをc-kitのリガンドであるSCF(stem cell factor)に置換した融合物(SCF-PE40)は、c-kit陽性肺小細胞癌(SY株)に特異的殺効果(障害)が観察された。
5.c-kit陰性乳癌細胞株(NCF-7)にc-kitcDNAトランスフェクションを行なった。本実験より得られたクローンの解析より、乳癌細胞増殖性に対するc-kitの抑制効果(負の制御)の一端が実施された。
