HTLV-1による遺伝子発現の活性化と抑制の研究

研究概要	HTLV-1はATLの原因となるレトロウイルスである。HTLV-1によるATLの発症機構について引き続き解析した。昨年度までの研究では、HTLV-1の制御因子Taxは転写活性化因子であり、リンホカインやその受容体、プロトオンコジーンなどの遺伝子の転写を活性化すること、それらの転写活性化が、TaxのNFkBへの結合による活性化、及びTaxのIkBへの結合による不安定化と、それに続くNFkBの核移行に伴う活性化により行われることを明らかにした。今年度は、Taxが細胞周期の阻害因子p16に結合し、その阻害活性を抑制する結果Cdk4キナーゼを活性化することを見出した。p16はCdk4に結合してキナーゼ活性を阻害し、その結果としてがん抑制遺伝子であるRb蛋白のリン酸化を抑制し、細胞周期をG1期に停止させる。この細胞周期阻害因子p16にTaxが結合することにより、G1期に停止した細胞が増殖を開始することも見出した。この知見は、転写活性化因子として作用するTaxが、直接細胞周期を調節する因子に結合することにより、細胞の異常増殖を誘導できることを示したものである。DNAウイルスが、がん抑制遺伝子Rbの機能阻害を通して発がんに関わることは既に知られているが、がん抑制遺伝Rbの機能阻害を通して発がんに関わることは既に知られているが、レトロウイルスであるHTLV-1の制御因子がRbとは異なる標的分子を介して同じRbによるシグナル伝達経路を阻害し、細胞の異常増殖に関わると言うこの発見は、ウイルス発がんの機構解析に与えるインパクトは大きい。このような状況から、Taxと16の相互作用がATLの発症に重要な役割を果たすことが示唆される。
