研究概要	本年度は、癌の本態解明に伴う癌治療の分子標的を意識しつつ以下の5つの課題に取り組んだ。 1.多剤耐性の分子機構と診断 2.抗癌剤誘導耐性の分子機構と治療への応用 3.アポトーシス誘導と耐性化の分子機構と治療への応用 4.抗癌剤の新しいターゲットの解明 5.転移癌細胞の特性の解明とその制御。本年の主要な成果として、多剤耐性、MMC耐性を克服する薬剤及びアポトーシス耐性について記述する。 MS-209は1〜10μMにおいてin vitroで多剤耐性を完全に克服する。耐性細胞移植マウスにおいても、抗癌剤との併用によりT/C150-200%の強い治療効果を示す。MS-209は健常人のphase I clinicaltrialで優れた薬理動態を示し、現在、leukemia、lymphoma、breast caを対象に臨床研究を開始した。KW2149はDT-diaphoraseが欠損したMMC耐性癌に有効な薬剤である。KW2149は生体内SHによって活性化されることが明かとなり、新しい薬剤創製に示唆を与えた。我々はU937細胞から抗癌剤VP-16によって誘導されるアポトーシスに抵抗性の株UK711の分離に成功した。UK711はVP-16誘導のアポトーシスのみならず、シトシンアラビノシド、アドリアマイシン、マイトマイシン、カンプトテシン、スタウロスポリン、サイクロヘキシミド、UVによるアポトーシスにも抵抗性を示し、これら薬剤によるcytotoxicityにも抵抗性を示すが、ビンクリスチン、TNF誘導のアポトーシスには抵抗性を示さない。UK711に代表される変異株はアポトーシスとの関連における薬剤抵抗性機序の研究に有用と思われる。
