細胞障害性リンパ球による標的細胞の障害機序

研究概要

パ-フォリンを発現していないCD4陽性T細胞クローン(BK1)による標的細胞障害に、標的細胞上のFas分子を介したアポトーシスの誘導が関与していることを見い出し、T細胞レセプターを介した刺激によるBK1上にFasに対するリガンド(FasL)が一過性に発現することを示した。また、RAG-2欠損マウスを用いたblastocyst complementation法により得られたパ-フォリン欠損T細胞から樹立したCD8陽性T細胞株(POK)においても同様なFasを介した細胞障害性が認められた。このようなFas依存性の細胞障害性(FasLの発現)は、これらの細胞株のみならず、一般のマウス脾臓由来のCD4陽性あるいはCD8陽性T細胞に広く認められたが、機能的FasLを欠損しているgld変異マウス由来のT細胞には認められなかった。以上の結果から、活性化T細胞上に発現するFasLは、パ-フォリン非依存性の標的細胞障害を媒介する主要な細胞障害因子であることが明らかとなった。
