研究概要

(1)ヒトARF依存性PLDのスプライシングバリアント(PLD1a,1b)を示し、PLD2の遺伝子をクロニーングした。ARF依存性PLDの増殖・分化の関連性を示唆した(野澤):(2)PAF-アセチルハイドロラーゼのI型は触媒サブユニット(α1,α2)とβサブユニット(Miller-Dieker症候群の原因遺伝子(LIS-1)産物)からなり、X線構造解析により三量体G蛋白質との類似性を示した(井上):(3)Ash/Grb2の下流分子をクローニングし、150kDaのPIP_2phosphataseを同定した(竹縄):(4)PAF受容体遺伝子のスプライシングバリアント(白血球型と組織特異的発現型)は異なるプロモーターに支配されていた。PAF受容体の過剰発現や欠損マウスで生理作用を検討した(清水):(5)ラット小腸ホスホリパーゼB/リパーゼの酵素活性中心を特定し、Gly-Asp-Ser-Leuモチーフに活性中心触媒基Serを持つ新しい型のSer-エステラーゼであった(岡本):(6)アミノペプチダーゼNの遺伝子クローニングのため、N末端及び内部アミノ酸配列を決定した。また、古細菌SulfolobusにGPIアンカー蛋白質が存在することを示した(池沢):(7)ホスファチジン酸ホスファターゼ(PAP)の2種類(a,b)のヒト2型PAPをクロニングした。PAP2aはwunenのホモログであり、PAP2bはDri遺伝子と同じものであった(加納):(8)マクロファージ分化抗原CD14がエンドトキシン(LPS)受容体であり、CD14を介するLPS刺激伝達にPLDが必須である(西島):(9)ラット腹水肝癌細胞核のPLA_2はII型PLA_2で、核に特異的に多く存在する(小泉):(10)エンドサイトーシス活性がPSフリッパーゼ不活性変異株で低下し、PSのゴルジ体への輸送にラット脳サイトソ-ルが関与することを示した(大木):(11)モルモット血漿PAF-アセチルハイドロラーゼのcDNAを単離し、酵素分子の一次構造を示した(野島)。
