難治性疾患に対する遺伝子治療の臨床への展開

研究概要	分子病態学的側面が明らかにされているが効果的な治療法のない難治性疾患に対して遺伝子治療が期待されているが、わが国での発展は欧米のそれに較べて遅い。我が国の将来的医療の発展を考えた場合この遅れは多くの禍根を残すことになる。そこで医学的必然性を誰もが納得できる形で示すために本邦での実態調査研究を行った。まず我が国における遺伝子治療のための基盤技術研究では、固形がん、悪性リンパ腫/白血病、単一遺伝子病(造血幹細胞を中心としたもの)、単一遺伝子病(それ以外の細胞を標的としたもの)、ウイルス性疾患ならびにその他(臓器移植とトレランスの誘導/臓器不全)の各項目について本邦ならびに世界における現況の解析がおこなわれた。これにより、各領域において我が国の遺伝子治療開発が急務であると考えられる疾患が浮き彫りにされてきた。つぎに全国の遺伝子研究関連施設研究者にたいして、遺伝子治療開始に対しての準備状況について15項目の153名の研究者を対象にアンケート調査を行った。回答率は63、3%であった。この結果では各研究者ならびに研究施設において遺伝子治療に対して積極的な取組みと方向性が明かとなった。現在、数施設において遺伝子治療臨床プロトコールの倫理審査が開始されているが、これらの遺伝子治療を適正に遂行していくために大いに参考になるものと考えられるので、今後引続き調査を行う予定である。
