光化学系II反応中心の分子構築と機能発現

研究概要	光化学系II反応中心の分子構築とその動態について解析し、以下の成果を得た。 (1)分子構築結晶化による構造解析を目標に、好熱性ラン藻Mastigocladusより光化学系II複合体を単離する方法を開発した。また、光化学系II反応中心のD1,D2タンパク質間の近接関係を架橋剤を用いて解析し、接触部位の存在をアミノ酸レベルで推定した。 (2)分子論的な動態暗冷所でのHPLC分析の手法を用い、単離した光化学系II反応中心に含まれるβ-カロチンが15シス型で存在することを明らかにした。また、蛍光励起スペクトルの解析から、光化学系II反応中心に含まれる不活性型(非反応型)フェオフィチンの同定に成功した。 (3)生物学的な動態 (1)光化学系II反応中心のD1サブユニットのC-末プロセシングに関与する酵素の純化に成功し、その遺伝子の同定と塩基配列の決定を行った。また、単離したプロテアーゼとD1タンパク質のC-末に相当するオリゴペプチドを用いて、酵素の基質認識機構を解析した。 (2)D1タンパク質の翻訳がポリペプチド伸長の特定の段階で光により制御されていることを見出し、その機構を解析した。 (3)D1タンパク質をコードする遺伝子(psbA)の特定の部位にin vitroでランダムな変異を入れ、強光下でスクリーニングすることにより、数種の光耐性シアノバクテリア(Synechocystis PCC 6803株)を得、その耐性機構を解析した。
