病原菌シグナル伝達と植物防御遺伝子応答におけるブラックボックスの分子生物学的解明

研究概要

エンドウ褐紋病菌のエリシターやサプレッサーは無傷エンドウ葉に対しても認識されたことより初期防御応答における細胞壁の重要性が示された。無傷エンドウ葉のエリシター処理による活性酸素種の生成、分離細胞壁画分のエリシター処理による感染阻害物質の生成はサプレッサーによって宿主特異的に阻害された。また、エリシター活性を保有するレクチン、コンカナバリンAの結合タンパク質複合体を原形質膜画分から単離すると、本複合体には防御応答に必須なポリホスホイノシチド代謝系のフォスファチジルイノシトールキナーゼや原形質膜ATPase、低分子量GTP結合タンパク質、ヌクレオシドジフォスフェイトキナーゼが存在することが明らかになった。これらのことから、エリシター・サプレッサーの受容体は細胞膜から細胞壁に存在していると考えられた。またポリホスホイノシチド代謝系の1産物であるジアシルグリセロールがエンドウ防御応答のセカンドメッセンジャーの候補であること、プロテインキナーゼCの活性がエリシター処理後数分以内に上昇すること、情報伝達におけるアクチン繊維の重要性が判明した。
フェニルアラニンアンモニアリアーゼやカルコン合成酵素などの防御応答遺伝子のプロモーターの詳細な解析は、エリシター応答には少なくとも複数種のシスエレメントが存在しており、このうちフェニルプロパノイド代謝系酵素遺伝子に共通して保存されている、Box-1,Box-2,G-Boxへの塩基置換の導入はレポーター遺伝子の発現を顕著に低減させること、更にその上流域にエンハンサー様配列が存在していることを明らかにした。また、トランジェントアッセイの系においてもサプレッサーはレポーター遺伝子の発現を顕著に抑えたことより、このプロモーター領域にはサプレッサーにも応答する領域が存在していることが明らかになった。
