研究概要	ウイルスの持続感染はウイルス側としては生体防御を回避し、生体内に存続する一つの寄生戦略として、また宿主側としては寄生体の排除よりむしろ共存を図る高度な戦略として捉えられる。本研究はネコ、イヌ、ニワトリで臨床上重要なネコ免疫不全ウイルス(FIV)、ネコヘルペスウイルス1型(FHV-1),イヌジステンパーウイルス(CDV)、ニワトリのマレック病ウイルス(MDV)による持続感染と再活性化の機構を培養細胞及び自然宿主を用いて、主として分子生物学的手法によって分子レベル、細胞レベル及び個体レベルを解明することを目的とする。本年度は以下のような成績を得た。 1)FIV感染猫のPBMC内で潜伏状態にあるFIVは調節遺伝子をコードしているmultiple-spliceのRNAを高率に発現するが、構造遺伝子やvif遺伝子をコードしている非spliceおよびsingle spliceのRNAの発現はほとんどない。しかし、PBMCをマイトジェンで刺激すると後者RNAは発現し、ウイルス産生が促進された。このことは潜伏状態のFIVは、その転写物の発現パターンを調節して、ウイルス産生を抑制していると考えられる。又FIVの潜伏状態から再活性化には調節遺伝子をコードしている転写物の発現バランスが重要と思われる。 2)FIV遺伝子発現の活性化および抑制にはLTRに存在するC/EBP結合部位がpositiveとnegativeの両方に関与することが示された。 3)In vitroにおけるFIV潜伏感染系をFIVを接種した人由来Molt4細胞を用いて確立した。この細胞において、FIVゲノムは感染初期に潜伏状態となり、TPA処理により活性化し、ウイルスを産生することが明かとなった。
