研究概要

ウイルスの持続感染はウイルス側としては生体防御を回避し、生体内に存続する一つの寄生戦略として、また宿主側としては寄生体の排除よりむしろ共存を図る高度な戦略として捉えられる。本研究はネコ、イヌ、ニワトリで臨床上重要なネコ免疫不全ウイルス(FIV)、ネコヘルペスウイルス1型(FHV-1)、イヌジステンパーウイルス(CDV)、ニワトリのマレック病ウイルス(MDV)による持続感染と再活性化の機構を培養細胞及び自然宿主を用いて、主として分子生物学的手法によって分子レベル、細胞レベル及び固体レベルで解明することを目的とする。本年度は以下のような成績を得た。
1.FIVのサブタイプA(Petalumaの株)およびサブタイプB(TM2株)のそれぞれを接種したSPF猫を約9年間観察した。前者は感染後8年4ケ月で腹腔内出血で死亡したが、後者は現在でも(9年以上)生存している。この結果、FIVの持続感染の場合、発病を左右する因子としてはウイルスのサブタイプ間の病原性の強弱並びに飼育環境下の差(例えば他の病原体に暴露されない場所)などが重要であると考えられた。
2.MDVは血清学的、遺伝学的に3つに分けられ、全て持続感染する。MDV-1は腫瘍原性を有するのに対して、MDV-2は非腫瘍原性である。持続感染並びに腫瘍原性の機構を遺伝子レベルで明らかにするために、MDV-2遺伝子を解析し、その90%以上を決定した。
3.ワクチン接種済みの犬からワクチン免疫から回避するCDVの野外株を分離し、そのH及びF遺伝子の解析を行った。その結果、塩基配列から予測されるアミノ酸解析において、野外株におけるH蛋白の異変がF蛋白より多いことが明らかとなった。
