慢性炎症性増殖性疾患の病態分子生物学的研究

研究概要	慢性炎症性増殖性疾患である慢性関節リウマチ(RA)の患者におけるBST-1の発現を検討するために、可溶性BST-1を特異的に測定できるELISAを樹立した。ELISAを用いてRAの患者血清中のBST-1を測定したところ、約7%の患者において、正常人に比較して約20倍の可溶型BST-1が存在することを明らかにした。可溶型BST-1は、大関節が破壊されるタイプのRAにおいて高値を示した。一方、RAにおけるBST-1の発現異常の機構を解明するために、BST-1の染色体遺伝子をクローニングした。現在、発現制御領域を解析中である。BST-1遺伝子の異常発現と免疫異常の関係を明らかにするために、BST-1トランスジェニックマウスを作製し、現在引き続き解析中である。又、BST-1遺伝子欠損マウスを作製する目的で、マウスのBST-1cDNAをクローニングし、その構造を決定した。引き続きマウスBST-1の染色体遺伝子をクローニングし、ジーンターゲッティングを行なうためのDNAを構築し、現在ES細胞にDNAを移入し、相同組み換えを行なっている。又、BST-1はCD38や、アメフラシのADPリボシールシクラーゼと一次構造が30%位類似しているが、ADPリボシールシクラーゼ活性があることを明らかにした。引き続き、本酵素活性とBST-1の生物活性の関係を明らかにする努力を行なっている。このように慢性炎症疾患のモデルであるRAにおいて、異常発現している遺伝子の一つであるBST-1を用いて、その異常発現の機構と、RAの病因、あるいは病態との関連を明らかにすべく研究を行なっている。一方、IL-6受容体を介するシグナル伝達機構に関しても、以下の点を明らかにした。1)junB遺伝子のIL-6応答領域にSTAT3が作用していること。2)Jakl,Jak2チロシンキナーゼや、FesチロシンキナーゼがIL-6受容体を介するシグナル伝達経路に関与していること。今後も引き続きIL-6受容体を介するシグナル伝達経路の解明と、炎症の関わりを明らかにしてゆく予定である。
