慢性炎症性増殖性疾患の病態分子生物学的研究

研究概要	慢性関節リウマチ(RA)患者の重症例で、血清中の可溶型BST-1が高値を示すことを明らかにした。さらに、可溶型BST-1の生体における役割を明らかにする目的で、可溶型BST-1を産生するトランスジェニックマウスを作製した。これらのトランスジェニックマウスには、免疫担当細胞の発生には特に障害は認められなかった。引き続き、免疫応答や、コラーゲン関節炎発症に及ぼす影響を検討中である。一方、BST-1の染色体遺伝子の構造をほぼ決定した。又、BST-1は、マウスの未熟Bリンパ球と未熟Tリンパ球において、共に抗原受容体遺伝子が再構成される時期に一致して一過性に発現されることを明らかにした。興味あることに、BST-1分子を抗体で架橋することによって、細胞内分子のチロシンリン酸化が誘導されることが判明した。このことは、BST-1は、受容体として機能し、細胞内になんらかのシグナルを伝達していることを示している。一方、IL-6受容体を介して、JAK-STATシグナルが活性化されるが、このシグナル伝達路は、IL-6によるマクロファージへの分化に必須であることを明らかにした。さらに、IL-6によって、BST-1の発現がマクロファージに誘導されることを明らかにした。これらの結果は、慢性炎症性疾患の局所において、IL-6とBST-1が、マクロファージの機能や分化を制御することによって、病態形成に関与していることを示唆しており、大変興味深い。
