研究概要	急性骨髄性白血病細胞および慢性骨髄性白血病急性転化期の骨髄芽球性白血病細胞の生物学的特徴である分化停止機構に着目し、その分子生物学的病因検索を行うことで新たな治療法開発に結びつけていく研究をいくつか行ってきた。この中でもまずG-CSFに特異的に反応して発現するGIG-1遺伝子のクローン化が慢性白血病患者白血病細胞から行われ、今後顆粒細胞の分化ならびにその停止機構の解析に重要な役割を演じるものと期待された。さらに分化停止機構の責任遺伝子の検討を行う目的でPEBP2αとPEBP2β遺伝子に注目し、研究を行った。16番染色体逆位異常をもつ急性骨髄性白血病細胞の切断点に位置する遺伝子、PEBP2β遺伝子はDNA非結合型の転写因子であり、DNA結合能のあるPEBP2αとヘテロダイマーを形成することが知られている。今回PEBP2β遺伝子のノックアウトマウスを作製したところ、そのホモ接合体で、明らかな胎児肝臓造血の抑制が観察された。ここでは骨髄球系細胞の分化抑制が観察され、これはAML1遺伝子にコードされるPEBP2αのノックアウトマウスで見られる造血抑制と同様な所見であった。このようにPEBP2αならびにPEBP2βの両遺伝子の白血病細胞分化停止機構への強い関与が明らかにされた。さらにEvi-1を含めたその他の転写因子と白血病化の関連、細胞周期関連遺伝子と白血病化との関連も現在検討を続けており、これらの解析結果は今後の白血病治療法への新たな展開をもたらすものと思われた。
