ヒト唾液腺癌細胞における新規な薬物受容体の検出と分化・アポトーシス誘導機構

研究概要	1.5-fluorodeoxyuridine monophosphate(FdUMP)受容体遺伝子を、ヒト唾液腺癌細胞(HSG)より単離し、このFdUMP受容体に対するcDNAを大腸菌にて発現させ受容体タンパクを調製し、FdUMP受容体に対する単クローン抗体を作製した。その結果、HSG細胞のFdUMPによる筋上皮細胞への分化とアポトーシスの誘導を阻止する単クローン抗体を得た。この場合、筋上皮細胞のマーカーとして、ミオシン、S-100タンパクβ鎖、飲小胞、ミオフィラメントの発現を蛍光抗体法、immunoblotting,電顕にて検索した。アポトーシスの誘導は電顕による核の凝縮、アガロース電気泳動法による核DNAの断裂、細胞DNAの3´-OH nick-end labellingにより検索した。 2.ヒト唾液腺腺扁平上皮癌細胞(TYS)をFdUMPで処理すると著明な増殖抑制の惹起されることを明らかにした。そこで、増殖抑制されたTYS細胞に発現されている遺伝子をクローニングした結果、FdUMP受容体遺伝子であることを示唆する遺伝子を得た。現在、この遺伝子についてさらに解析を行っている。 3.FdUMP受容体は、^3H標識した5-fluorouracilとそのアナログを用いたカラムクロマトグラフィーによる解析により、Fに特異的な受容体である可能性が示唆された。 4.FdUMP受容体を発現していないヒト癌細胞(唾液腺或いは口腔粘膜)に、FdUMP受容体をコードしているcDNAをtransfectionしてFdUMP受容体を発現させた。この細胞において、FdUMPによりアポトーシスの誘導の起こることを示唆する実験結果を得た。
