ヒト唾液腺癌細胞における新規な薬物受容体の検出と分化・アポトーシス誘導機構

研究概要

1.CNBr-activated Sepharose 4Bに5-fluorodeoxyuridine monophosphate(FdUMP)をカップリングさせ、FdUMPアフィニティカラムを作製し、HSG細胞より調製したタンパク抽出液を結合させ、溶出緩衝液で結合タンパクを溶出し、分子量約28kDaのタンパクを得た。このタンパクのアミノ酸シークエンスの分析の結果、NLDLDXIXXXVKAという9残基のアミノ酸が特定された。アミノ酸残基のホモロジーサーチにより、type II keratinであるCK5,CK6,CK8の配列であるNLDLDSIIAEVKAと解読できこのアミノ酸残基は100%一致し、未解読の部分もその配列は完全に一致した。以上の所見は、FdUMPはサイトケラチンと直接結合するか、または中間径フィラメントに結合しているアダプター分子と結合することを示唆している。
2.HSG細胞よりRNAを抽出して精製したmRNAを用いて、oligo dTプライマとrandomプライマを用いて逆転写を行いcDNAを調製して、λZAP Expressファージベクターにプロモーターに対してアンチセンス方向に挿入し、anti-sense oriented cDNA libraryを構築した。この発現ライブラリーをHSG細胞にトランスフェクションして、FdUMP存在下でも増殖可能な8個の細胞クローンを分離した。このクローンよりcDNA断片を回収し、塩基配列を決定した。その結果、プロテオソームを構成する酵素の遺伝子とプレクチン遺伝子を検出した。プレクチンは、中間径フィラメント、MAP1と2、細胞膜骨格タンパク、ラミンBと結合し、細胞膜と核膜をつなぐアダプター分子であると考えられている。以上の結果より、FdUMP結合タンパクとして精製されたII型ケラチンは、プレクチンを介してFdUMPと結合し、FdUMPのHSG細胞の増殖抑制と分化あるいはアポトーシス誘導に関連したシグナルを伝達しているものと考えられる。更にその下流にプロテオソームによるある種の蛋白分会酵素がFdUMPの細胞増殖抑制シグナルにおいて重要な役割を担っているものと思われる。
