研究概要	バクテリオロドプシンにおいて、そのプロトンポンプの最も重要なステップである、光吸収後のシッフ塩基からAsp85へののプロトン移動を起こすL中間体の構造を、フーリエ変換赤外吸収スペクトル法で解明した。 (1)Asp212をアスパラギンに変えた変異蛋白質では、シッフ塩基からAsp85へののプロトンの移動が可能な形になっているにも関わらず、実際にはプロトンの移動はない。この変異蛋白質のL中間体のFTIRを測定すると、野性型でみられたAsp85のそばにある水分子の強い水素結合の形成が見られない。水分子の作るAsp212、Asp85、シッフ塩基との強い水素結合が、シッフ塩基からAsp85へのプロトンの移動に必須であることが結論された。また、Val49、Ala53などリジン側鎖の近くにある残基を変えるとやはり水分子の強い水素結合の形成がなくなると共にプロトンの移動が抑えられた。 (2)野性型の9位のメチル基をなくしたバクテリオロドプシン、およびTrp182の変異蛋白質をつかい、L中間体ではTrp182のインドールのN-Hは、9メチル基を介してレチナ-ルのポリエンと相互作用し、シッフ塩基からAsp85へのプロトンの移動を促進していることを明らかにした。 (3)L中間体ではシッフ塩基はさらに、その細胞外側に連なったAsp85から遠く離れた、細胞質側のThr46、Asp96の近くにある2分子の水の水素結合を変化させることを示した。 (4)ナノ秒段階の反応を解析し、K中間体が約10ナノ秒の寿命で次のKL中間体に変換すること、この二つの中間体は、シッフ塩基の近くの捻れ方にあることを示した。
