ネコ・サルにみられる四足および二足歩行運動の高次制御機序

研究概要	1.ネコの四足歩行運動とその集中制御機序本研究から除脳ネコ歩行標本の小脳白質部に、その部位の連続微小電気刺激で四足歩行運動が誘発できる小脳歩行誘発野を新しく同定することができた。 1)小脳歩行誘発野と網様体脊髄路細胞小脳歩行誘発野は左右の室頂核遠心路線維が交叉する小脳白質の正中部に相当した。この部位に微小電気刺激を加え、網様体脊髄路細胞におよぼすその刺激効果をシステマティックに解析した。その結果、それらの細胞は小脳白質正中部から巣シナプス性の促通効果をうけること、さらに網様体脊髄路線維の中でも脊髄白質の腹内側部を下行する線維が歩行運動の発動信号を伝達していることが解明できた。 2)網様体脊髄細胞と脊髄介在細胞網様体脊髄細胞が局在する橋・延髄網様核に微小電気刺激を加え、腰髄レベルにおいて順行性に興奮性シナプス接続をうける介在細胞を軸索内微小記録を用いて同定した。これら介在細胞の中でも中脳歩行誘発野の刺激で律動性発射活動を示す軸索の細胞体は脊髄灰白質の第VIII層に局在し、それらはその軸索を細胞体の位置として反対側の脊髄白質部に投射していることを明らかにできた。 2.サルの直立二足歩行運動とその高次制御機序流れベルト上での直立二足歩行運動を学習・習得した成サルはかかと部から着地するヒト型の歩行様式を示した。これらのサルの頭蓋部を部分開頭し、大脳皮質の補足運動野、運動・感覚野を中心としてそれぞれの部位に微小電気刺激を加え、後肢運動が発現する部位のシステマティックなマッピングを行った。歩行運動を学習したサルでは未学習なサルに比べて大脳皮質の後肢支配領域が機能的に変化していることを示唆する実験成績が得られた。この成績は直立二足歩行運動の学習に大脳皮質が直接的に関与していることを推定させ、その推定を検証することが今後の重要な研究課題となると考えられる。
