蛋白脱リン酸化酵素阻害剤トートマイシンの作用機構に関する化学的研究

研究概要	1)分子レベルでの研究を進める場合、トートマイシンの化学構造を正しく決定する事は最重要課題である。トートマイシンより得られるフラグメントの化学合成に成功して、不明であったトートマイシンの部分構造を確定した。 2)トートマイシンから逆合成した3つのセグエントA、B、Cの立体選択的合成に成功した。無水マレイン酸骨格を有するセグメントAの合成はフランの光増感酸素酸化を用いることによって達成した。アミノ酸D-バリンを出発原料として、立体選択液四酸化オスミウム酸化反応を用いてセグメントBの有する3つの不斉中心を構築した。そしてジチアンアニオンとエポキシドとのカップリング反応を用いてセグメントBの合成を完了した。ピラノース糖を出発原料とし、コバルト錯体を用いて疑エナンチオ選択的ヘテロ共役付加反応を開発して、セグメントCの立体制御合成を行った。 3)スルオンカルバニオンとエポキシドの反応、改良山口法を用いたエステル化反応によってセグメントA、B、Cをカップリングしてトートマイシンの有するすべての炭素骨格と不斉中心が揃った化合物を得た。無水マレイン酸部分の構築と脱保護を行い、トートマイシンの全合成を完了した。 4)分離力場計算と超伝導核磁気共鳴装置による立体配座解析を行い、溶液中における立体化学を決定した。 5)合成中間体を用いて、トートマイシンの誘導体を合成して、阻害活性の評価を行った。そして阻害活性発現に必要となる官能基を決定した。
