研究概要

糖鎖の機能を探る一つの手法として、糖転移酵素の選択的阻害による糖鎖合成の制御の可能性の追求を、グリコシド結合の加水分解酵素(グリコシダーゼ)および形成酵素(トランスフェラーゼ)の阻害剤のデザインを通して行っている。グリコシダーゼ阻害剤では、従来から進めてきた5-チオアルドース、とくに5-チオ-L-フコースを含むオリゴ糖と、そのフコシダーゼに対する阻害活性を集大成し、グリコシド結合様式による相違が見られることを明らかにした。さらに、より強い活性の期待されるアザ糖を非還元末端に有する擬似オリゴ糖へ展開するため、非環状誘導体の形で、環元末端側の糖を導入したのちに、アザ糖ピラノシドを形成する方法を開発した。糖転移酵素としてはスクロースから異なったO-グリコシルフルクトースを形成するグリコシルトランスフェラーゼおよびβ1→4ガラクトシルトランスフェラーゼに対して生物有機化学的手法により基質認識を明らかにした。前者では種々の置換スクロースを、後者では種々の供与体(UDP-ガラクトース)のアナログを合成し、それらを基質とした転移反応より、酵素の活性部位における分子認識を探る手がかりとなる知見を得た。
