研究概要

5-チオ-L-フコースを含むオリゴ糖の合成においては、従来のフコシダーゼによる結合様式特異的な認識を検証するプローブとしてのみならず、レクチンや抗体による糖鎖認識を解明するプローブの開発へと展開した。すなわち、血液型関連抗原であるH type2およびLe^x三糖のフコース残基を5-チオフコースに置換した三糖アナログを、共通の合成中間体である1,6-アンヒドロ-2-アジド-2-デオキシ-4-O-(β-ガラクトシル)グルコース誘導体を経由して合成した。合成したH type2三糖アナログは、H type2三糖を認識するレクチンに認識されず、抗体に強く認識されるという、その普遍性に関して今後の検討に興味のもたれる事実を見い出した。
アザ糖を含むオリゴ糖の合成においては、アザ糖を他の糖に結合する構造として分子間アミジン結合に着目し、チオアミドとアミンから水銀塩の存在下に構築する効率の良い方法を見い出した。実際の二糖アナログの合成にあたってはアミジンの形成を結合の分子間または分子内で行う2つのルートを開発した。これによりエンドグリコシダーゼの特異的阻害剤を合成しうる可能性を見い出した。また、合成スクロースアナログを用いて、2種の微生物由来のα-グルコシルトランスフェラーゼの基質特異性の発現に関与する水酸基を明らかにした。
