レトロポゾンの進化に関する分子集団遺伝学的研究

研究概要	より広い種でのIエレメントの進化を明らかにするために、キイロショウジョウバエと同じSophophola亜属のmontium種亜群5種、他の亜属であるScaptodrosophila亜属1種、Drosophila亜属2種、鱗翅目のカイコ、計9種のゲノミックライブラリを作成し、キイロショウジョウバエのIエレメントにホモロジーのあるエレメントのクローニングを行い、それらの構造、染色体上の分布等を決定し、分子進化学的な解析をおこなった。montium種亜群とScaptodrosophila亜属のD.lebanonensisからはデータベースサーチの結果から、Iエレメントであることが確かめられたエレメントがいくつか得られた。それらはキイロショウジョウバエの完全型のエレメントと塩基配列はかなり異なっていたが、gagや逆転写酵素の領域、およびTAAリピートが保存されているものもみられた。その系統関係はhost speciesの系統関係と同様であり、これらの種の種分化以前に挿入が起こったと考えられる。キイロショウジョウバエ種亜群で見られた水平感染の例は見つからなかった。Iエレメント相同配列は、染色体上にはキイロショウジョウバエ種亜群同様に染色体腕上と染色中心に存在しており、とくに染色中心に多くみられた。LINE-likeエレメントは数多くの生物種に存在しており、その系統関係からも、それらの起源はかなり古いと考えられる。この研究は、ショウジョウバエのIエレメントをモデルとして、レトロポゾンの進化の様相とその機構を明らかにする目的で行われた。Iエレメントのショウジョウバエにおける分子進化の様相を知り、また単純なモデルを使ったモデル解析には成果を上げることが出来たが、その進化機構についてはまだまだ実験的、理論的には解明されなければならない問題が多くあり、さらに研究を続けていく必要がある。
