研究概要	1.ウナギの心臓を単離して灌流標本を作成し、灌流液の浸透圧を上昇させると、心房性及び心室性ナトリウム利尿プペチド(ANP、VNP)の分泌が上昇した。従って、心筋細胞自身が浸透圧受容体として機能していることがわかった。心房と心室を分けて細切し、イオン交換ゲルに埋め込んで表面を灌流したところ、心房からはANPとVNPが、心室からはVNPのみが分泌されていることがわかった。また、ANPは成熟型が分泌され、VNPはプロホルモンが分泌されていた。 2.放射性リガンド結合実験を用いて鰓のANP受容体の性質を調べたところ、親和性は血漿ANP濃度とほぼ等しく、ほとんどの受容体はC型であることがわかった。しかし、淡水ウナギの鰓ではC型ナトリウム利尿プペチド(CNP)がとりわけcGMP産生を増加させたため、少量のB型受容体も存在することがわかった。 3.血圧に影響を与えない低用量のANPをウナギの血液中に継時注入したところ、尿量は減少して尿中のナトリウム、カルシウム、マグネシウムの排泄量が増加した。また、血漿ナトリウム濃度や浸透圧はANPの投与時に低下した。従って、ANPは鰓からのナトリウム排泄も増加させていると考えられる。 4.哺乳類と同様に、半水生の両生類であるウシガエル心室からB型ナトリウム利尿プペチド(BNP)を単離・同定した。従って、両生類は魚類型のVNPではなく、陸上動物型のBNPを持つことがわかった。
