ホヤのバナジウム濃縮のエネルギー機構と高選択性の解明

研究概要

原索動物のホヤは遷移金属元素のバナジウムを高選択的に濃縮していることでよく知られている。これまでバナジウムの濃縮に関与する物質としていくつかの低分子が挙げられてきたが、タンパク質を始めとする高分子の関与についてはほとんど報告がない。ホヤが海水中のバナジウムを高選択的に取り込み濃縮する際の経路を考えると、バナドサイトは濃縮経路の末端に位置する。バナジウムがこの細胞に濃縮されるまでには、エラを通った多くのイオンのなかからバナジウムのみを選択してバナドサイトの細胞膜まで運搬する物質(バナジウムトランスファー)、バナドサイトの細胞膜上でそれを受ける受容体(バナジウムレセプター)、そしてバナジウムイオンが貫通する膜チャンネル(バナジウムチャンネル)などが必要と思われる。われわれはこれまでに、高濃度のバナジウムを含有するアスキジア科のホヤ、Ascidia sydneiensis sameaの血球細胞から少なくとも4種類(約100kDa,45kDa,15kDa,12.5kDa)のバナジウム結合タンパク質を得ることに成功した。低分子量のそれについては、アミノ酸の部分配列の決定とモノクローナル抗体の作製に成功し、このタンパク質はバナドサイトの膜上に存在することとタンパク質1moleあたり約0.5moleのバナジウムが結合していることを見出した。
