研究概要	Phytophthora属菌の宿主選択的過敏感細胞反応抑制作用を示すサプレッサーグルカンの分離・精製ならびに作用軌溝の解析を次のようにすすめ、成果を得た。1)Phytophthora infestansの遊走子の透析膜上接種による膜侵入内液ならびにCa^<2+>添加浸透胞子発芽培養液より、フェノール処理、クロロフォルム・メタノール混液処理を経て、水溶性画分に回収されるグルカンを、陰イオン交換クロマトグラフィーにかけ、中性および4つの陰イオン性グルカンを分離した。侵入液中では全てが陰イオン性グルカンで、発芽液中では平均60%が陰イオン性グルカンであった。2)各分画のグルカンをアミドカラムHPLC分析した結果、いずれも18-23の重合度で、陰イオン性グルカンはグルカン分子当たり1-4分子のリン酸化されたグルカンと推定した。3)陰イオン性グルカンのSW3000カラムによるHPLc分析では、酸性条件下ではほぼ分子量に相当する1画分として分離されたが、弱アルカリ性溶出条件で4つに分画され、重合度が同じでもpHにより異なる分子的性質もつグルカンの存在が確認された。3)サプレッサーの過敏感反応抑制活性とその機構解析のため、菌体エリシター刺激による活性酸素生成の簡易測定法を工夫し、ルミノール化学発光を応用したオートフォトグラフィー法ならびにフォトカウンティング法と、蛍光指示薬を負荷した細胞蛍光のクエンチング測定法を確立し、サプレッサー活性検定を可能にした。4)膜近辺のCa^<2+>とキレートして蛍光化するクロロテトラサイクリンの蛍光追跡法を開発し、サプレッサーグルカンがエリシター刺激による活性酸素生成系の活性化を阻害し、これは刺激後1-2分以内におこる細胞外Ca^<2+>のチャンネルを介しての細胞内流入系を阻害する可能性を示した。5)プロトプラストが第一次侵入菌糸嚢と反応し過敏感反応を示し、その反応開始10秒前に侵入液グルカンを加えるとそれが抑制されることを示し、感染現場での機能性を示唆した。
