研究概要	前年度、Phytophthora属菌のフォスフォサプレッサーグルカン(P-GS)の作用機構の解析の結果、それが宿主組織の誘導抵抗性の引き金的存在の原形質膜局在の活性酸素生成系の活性化過程の細胞外Ca^<2+>の流入を阻害することを明らかにしたが、このことをさらに深めるため、定量的にCa^<2+>を結合させた原形質膜小胞を用い、疏水環境下でCa^<2+>との結合し蛍光化するクロロテトラサイクリン(CTC)の蛍光変化を指標に、菌体エリシターとP-GS処理による膜表面のCa^<2+>の動向を調査した。その結果、宿主原形質膜表面には一定量のCa^<2+>結合部位があり、エリシターはその結合を解除するが親和性菌由来のP-SGはその解除を阻害することが推察された。しかし、無細胞系の膜画分にはグルカナーゼやフォスファターゼ活性が存在し、P-SGを分解するため、その膜分子との結合実験にはさらに工夫のいることが判明した。構造解析については、分子量的に同じ分画のP-グルカンがpHによりクロマトグラフ的性質を変化させ、さらに分離し純化をさらにすすめた。個々の分離グルカンの活性検定を前年度開発したエリシター刺激の活性酸素生成反応の抑止作用で試みたが、宿主ならびにその品種に対して異なったサプレサ-活性を示すことが示唆され、構造の異なる多様なグルカンが菌より分泌されていることが判明した。そのため、構造解析には特定の分画に絞って量的に確保し検討することの必要性が生まれ、今後の検討課題とした。
