条虫類の宿主転換(野生動物⇔家畜⇔ヒト)と適応放散

研究概要

本年度は、これまで分離されたTaenia taeniaformis 10分離株ならびにEchinococcus multilocularis 6分離株を含むTaenia属7種とEchinococcus属2種についてCOI(mitocondorial cytochrome c oxidase subunit I)遺伝子(391bp)について比較した。これらのテニア科条虫の間で、12%程度の塩基配列の相違を示したものは独立種と考えられた。幼虫型(metacestode)で分離された多包条虫E.multilocularisでは動物種(エゾヤチネズミ、ツンドラハタネズミ、ドブネズミ、ブタ)ならびに地域(日本、アラスカ)に関わらず同一の塩基配列を示した。これに対して、分離された地域、中間宿主の異なる従来の形態学的特徴から猫条虫Taenia taeniaeformisとして位置づけられた10分離株について検討したところ北海道虻田のエゾヤチネズミからの分離株(TtACR)を除いて、0.3〜4.1%の範囲で相違が見られた。しかしながら、TtACR9.0〜9.5%の範囲で相違が見られ、独立株あるいは独立種のレベルの差が見られることが明らかになった。北海道には従来の猫条虫とは起源の異なるが、現存するネコに適応したテニア属条虫が存在することが明らかとなった。本分離株は形態的にも小鉤の特徴ならびに中間宿主の選択性(エゾヤチネズミに適応)などの生態的特徴からも独立種の可能性が高い。北海道から絶滅したネコ科動物から近年になって移入されたイエネコを代替宿主として維持されていた遺残種と考えられる。テニア属条虫の系統を論じる上で重要である。なお、虻田のエゾヤチネズミで分離されたのを最後にその後、野外から採取されていないが、実験室内で系統が保存されている。
