培養細胞と単離視床下部ニューロンを使った温度需要機構解析

研究概要

本研究は、従来皮膚の温度受容器について提唱されていた仮説や、細胞外記録による間接的証拠で説明されていた温度受容機構を、我々の最近の知見を基に、(1)末梢交感神経モデルとしての培養細胞PC-12による冷受容機構の解析、(2)体温調節中枢である視床下部視索前野の単離ニューロンを使った温度受容機構の解析、(3)温度受容機構におけるカルシウムイオンの役割解明の3方向から追求し、温度受容機構の基本的原理をイオンチャネルあるいは細胞レベルで解明することであり、以下の点を明らかにした。
1、PC-12細胞における冷受容機構の解析
未分化の、あるいは神経成長因子を作用させて分化させたPC-12細胞の温度応答をホールセルクランプ法あるいはグラミンジンパッチクランプ法で記録した。膜電位固定下に細胞外液温を30-39℃の温度範囲で変化させ、温度刺激応答時のシオン電流のkinetics解析と、分化の過程における温度応答性の変化とイオン電流との関連を調べると、未分化あるいは分化させたPC-12細胞のいづれの細胞においても、脱分極刺激時にみられる内向き電流の大きさが細胞温度の下降により増大し、冷感受性を示すが、その増大の程度は36-38℃で顕著であった。
2、温度刺激ときの細胞内カルシウム濃度の動態解析
蛍光性カルシウム指示薬fura-2をPC-12細胞に負荷し、温度刺激応答時の細胞内カルシウム濃度の動態を測定した。培養液温を34℃から変化させると、温度上昇、下降のいずれの場合も細胞内カルシウムは一過性に増加した。細胞内ストア-由来のこのカルシウムどのように温度受容機構に関与しているかを明らかにする予定である。
