研究概要

近年アラキドン酸遊離反応を担う酵素として、分子量85K細胞質ホスホリパーゼA_2(Cytosolic phospholipase A_2略してcPLA_2)が、低濃度カルシウム感受性で、アラキドン酸選択性であることから注目を集めている。
1.申請者は、培養線維芽細胞を腫瘍壊死因子で処理する事により誘導されてくるcPLA_2とは異なる新規のホスホリパーゼA_2を検出していた。このPLA_2は分子量60Kで、低濃度カルシウム感受性でアラキドン酸選択性のPLA_2であった。そこで、この酵素を誘導型PLA_2“Inductive phospholipase A_2"(略してiPLA_2)と名付け、以下の解析を行った。
エンドトキシンを投与したラット脳や肺にアラキドン酸選択的PLA_2が誘導された。陰イオン交換カラム、疎水カラム、ゲルろかカラム上での挙動は線維芽細胞で検出したiPLA_2と同様のパターンを示した。またその性状もカルシウム感受性やアラキドン酸選択性で同一であった。さらにエンドトキシン処理したウサギ脳でも同様の現象が検出された。このことより、このiPLA_2は種を越え、様々な臓器に存在しており、重要な生理的役割を担っている可能性があると考えられる。現在、ラット脳と肺よりこのiPLA_2の完全精製と構造決定を進めている。
2.cPLA_2はMAPキナーゼでリン酸化されることで、活性調節を行っている。大腸菌に発現させたcPLA_2タンパクを市販のヒトデMAPキナーゼでリン酸化し、リン酸化cPLA_2に反応するが、非リン酸化cPLA_2には反応しない単クローン抗体を調製した。これを用いて、新しい情報因子としてのcPLA_2の解析をスタートした。
