マスト細胞プロテアーゼmRNAの発現をマーカーとしたマスト細胞の多様性の解析

研究概要	マスト細胞の好塩基顆粒中には多くの種類のマスト細胞特異的なプロテアーゼが含まれている。マウスではプロテアーゼの遺伝子の多くクローニングされており、マウス・マスト細胞・プロテアーゼ(MMCP)の1から5までがキマ-ゼ、MMCP-6とMMCP-7はトリプターゼであることが知られている。おのおののMMCPの発現がいかなる要因により制御されているか調べてみると、まずマウスの系統によって変化し、同じ系統ではマウス細胞の存在する組織、サイトカイン受容体からのシグナル、転写因子などによって影響を受ける。本年度はサイトカイン受容体からのシグナルと転写因子のMMCP-6の発現に対する影響を調べた。c-kitはマスト細胞の最も重要な増殖因子stem cell factor(SCF)の受容体であるが、我々はc-kitの機能獲得性の突然変異を発見している。この突然変異c-kitのcDNAをMMCP-6を発現していないマスト細胞株IC-2に導入すると、MMCP-6のmRNAの発現は著明に増強した。またこのIC-2細胞におけるMMCP-6の発現は培養液にインターロイキン3を加えると再び減弱した。マウスのmi遺伝子座がコードする転写因子(MITF)はMMCP-6の発現に影響を与える。mi遺伝子座の突然変異遺伝子を2個持つmi/miマウスの皮膚のマスト細胞はMMCP-6をほとんど発現していない。mi/miマウスの脾細胞を培養して得たマスト細胞もMMCP-6を発現していないが、このmi/miマウス由来培養マストに正常にMITFのcDNAを導入するとMMCP-6のmRNAの発現は正常化した。この現象をin situハイブリダイゼーションで調べてみると、ノーザンブロットとよく一致する結果が得られた。
