研究概要

1.OPTMの作出:メタロチオネイン(MT)プロモーターをマウスのOP遺伝子の5′上流に連結した変異遺伝子を作成し、現在C57BL/6Nマウスのトランスジェニックマウス(TM)を作成し、C57BL/6NマウスにおけるMTプロモーターのTM(MTTM)をこれまで12系統作出した。これらの各系統のうち4系統で第2世代の出生が観察されたが、第3世代で妊性が見られないか胎生期あるいは新生仔の段階で全部淘汰されMTTMの維持に困難を極めた。第1および第2世代の3匹を屠殺したが、肉眼および光顕的に著変は見られなかった。そこで肝臓で強く発現するα1アンチトリプシン(α1AT)をプロモーターにし、OP部をOP cDNAを用いたTMの作成をF1マウスで試みた。これにより現在まで10系統のα1ATTMを作出することに成功した。現在これらのTMの繁殖を開始したところで、諸解析はこれからである。
