熱帯熱マラリア原虫の47および23kD分子の純化と防御抗原性に関する研究

研究概要

平成7年度において長年続けられた本研究はようやく最終段階に到達した。本研究の目標は、マラリア流行地から得た患者血清を(1)急性期で臨床症状を呈している患者からの血清(2)マラリアが慢性化し脾腫が顕著な患者で原虫に感染しながら臨床的に発症していない患者からの血清グループに分けて、おのおのの血清が特異的に反応する熱帯熱マラリア原虫分画抗原をWesternblot法により検出すること、さらに進んで検出された分子量のマラリア原虫抗原を純化された物質として一定量を用意すること、そしてその物質がマラリア抗原として防御に関与した免疫反応を生体に惹起させることができるかどうかをまずin vitroの系でしらべ、有望な結果がみられた場合ヨザルを使ってin vivoの実験を進め、マラリアワクチン候補抗原を特定すること、以上のような研究計画が立てられ実施されてきた。まず、(1)の血清を使い、急性マラリアを乗り切るときに47kDポリペプチドが、何らかの免疫に関与していることが判明したため、熱帯熱マラリア原虫からDNAをとりだし、cDNAあるいはgDNAのライブラリーを作った。用意されたライブラリーを使って47kDポリペプチドを作り出すことは、結果的に失敗に終わった。そこで今般はタンパク化学の技法を採用し大量培養された原虫から47kDポリペプチドを純化単離し、さらに得られた分画をaffinity chromatographyにより抗47モノクロナール抗体に結合する分子をゲル内に捕捉し余分な成分を洗い流したのちにelution bufferを使って回収した。このようにして得られた分子は現在、マラリア患者から採取した抗原提示細胞とリンパ球よりなる培養系に作用させて、in vitro系での免疫反応を確かめるべく現在保管されている。慢性マラリア状態を反映する23kDポリペプチドについては、Prp-cell econo-automation system (Bio Rad)により電気泳動分画された状態で保管されてある。
