T細胞受容体複合体シグナルモチーフを介する情報伝達機構の解析

研究概要

TCR-CD3複合体を介するシグナル伝達系において、CD3γδεとζとがin vivoでも機能的に異なるシグナル伝達を誘導するか調べるために、シグナルモチーフを欠損させたζ鎖のトランスジェニックマウスを作製し、ζ欠損マウスとかけあわせた変異ζマウスにおけるT細胞分化を解析した。ζ欠損マウスでは、未熟、成熟胸腺細胞の両者が分化阻害を受けたが、変異ζマウスでは、成熟細胞のみに分化の亢進がみられたことから、ζモチーフを介するシグナルが未熟細胞の分化・増殖に重要なことが判明した。このことから、in vivoでもCD3γδεとζとは異なるシグナルモジュールを形成していることが示唆された。
各々のシグナル経路に特異的な伝達分子をクローニングするため、CD3ε、ζの細胞内ドメインのGST融合分子を作製し、これを蛋白プローブとしてλgt11のT細胞cDNAライブラリーをスクリーニングした。リコンビナントLckを用いてζ、εをリン酸化し、リン酸化プローブを用いる試みも行ったが、効率の良いリン酸化は得られなかった。非リン酸プローブを用いて、CD3εに特異的に結合する分子を5個クローニングした。この内の1クローンは、トポイソメラーゼIIβのC末端側部分であった。
TopoIIβは、CD3ε,FcRγの細胞内領域とは結合するが、ζの細胞内領域とは結合せず、結合の特異性が存在した。TopoIIβ側は、C末端付近の活性調節領域付近に結合部位を同定した。CD3ε、ζが活性化に伴ってdetergent insoluble分画に移行すること、およびTopoIIβはαと異なり、分裂にともなって細胞質にも局在するなどが判明した。in vivoでの会合およびT細胞活性化にともなってCD3εが膜から移行し、核内蛋白と結合する可能性を含め、その意義を解析している。また他の4クローンについても塩基配列決定、結合特異性と共にin vivoでの会合を解析中である。
