T細胞受容体複合体シグナルモチーフを介する情報伝達機構の解析

研究概要	T細胞レセプター(TCR)・CD3複合体を介する抗原認識シグナルの伝達機構の解析のために、すでに明らかにした2つのシグナルユニットであるCD3ζとCD3εを介する新たなシグナル伝達分子を同定した。シグナルモチーフを有するζ鎖を介してチロシンリン酸化を誘導する90kd蛋白を同定した。この蛋白のリン酸化は、ζモチーフに依存し、TCR刺激でのみを誘導される新たな分子であると思われる。他方のユニットCD3εに結合する蛋白を2分子同定した。1つはトポイソメラーゼllβで、両者の結合領域を同定した。topollβは核内に局在しているが、我々はTCR活性化にともなってζ、εが核分画に存在し、topollβと会合することを初めて示した。もう1つの分子も、T細胞中でCD3εとのみ特異的に会合する未知の分子であることが判明した。一方、目的の1つであったリン酸化ζペプチドを用いた結合分子のスクリーニングは成功するにいたっていない。また、TCRシグナルによるT細胞増殖と文化の制御の解析を行った。増殖しているT細胞をTCR刺激すると細胞周期がG1で停止し増殖阻害が起こることがT細胞クローンでも確認された。これは、ハイブリドーマの場合と同様にサイクリンD3とそれに会合するキナーゼ活性が著しく抑制されるためであると思われた。増殖シグナルとの関係を解析した結果、TCRシグナルによってJak3が低下し、IL-2不応答性が誘導されている可能性を示唆した。一方、TCRシグナルの分化の制御については、雄抗原特異的TCRトランスジェニックとζ欠損マウスの交配マウスの解析を行い、TCRの発現レベルとシグナル伝達の強さを変化させることによって、ポジティブ・ネガティブ選択が変化しうることを初めて明かかにし、これまでペプチド濃度を変化させることによって示されてきたT細胞選択のアビディティーモデルを、T細胞の側から明らかにした。
