胸腺外T細胞初期分化

研究概要	(1)胸腺型と末梢型NKT細胞の相違結論:両者は異なる細胞集団である。理由:胸腺型NKT細胞は細胞表面にVβ8受容体のみを表現する。細胞質内にVα14Jα281mRNAをもつが表現していない。・一方、末梢型NKT細胞は2種類あり、Vβ8^+/Vα14^+型とVα14^+/Vβ8^+受容体発現型である。 (2)自己免疫病発症とNKT細胞の関係・自己免疫病発症前から末梢とNKT細胞は全体に減少する。・しかし、Vα14^-/Vβ8^+NKT細胞は残るが、Vα14^+/Vβ8^+NKT細胞は完全消失することからVα14NKT細胞と自己免疫病発症の相関が認められた。このことはVα14^+/Vβ8^+NKT細胞のリガンドは別々で二段階のセレクションがあることを示唆する。抗Vα14抗体の投与によって自己免疫病の早期発症をみたことから、Vα14NKT細胞が自己免疫病発症を制御していることが明らかとなった。・ヒトにおいてもSLE患者、強皮症患者においてマウスVα14Jα281と相同のVα24JαQの消失と自己免疫病の発症の相関がみられた。 (3)胎生初期分化・通常のT細胞分化と異なり、胎生9.5日AGM(Apotogenitomesonephton)領域から出現する極めて異例のT細胞であることも判明した。・もちろん胸腺形成前からT細胞であることも判明した。 (4)NKT細胞のリガンド Bendelacによって胸腺型Vα14^-/Vβ8^+NKT細胞のリガンドはCD1であることが判明したが、末梢NKTの大部分を占めるVα14^+NKT細胞のリガンドは別である可能性が判明した。しかし、リガンドの同定にまで至っていない。
