可溶性スーパー抗原の作成とそれによる免疫寛容の機構の解明と臨床応用

研究概要	(1)Mouse mammary tumor virus由来可溶性スーパー抗原の作成:ヒト免疫グロブリンIgGlの定常領域遺伝子とMtv7スーパー抗原遺伝子とを連結しpBOS vectorに挿入し、マウスB細胞腫瘍にトランスフェクションをした。このfusion proteinはELISA法により検出できるが、その機能は(V_β6^+T cellsの活性化)はつかまっていない。 (2)V_β4に特異的スーパー抗原の遺伝子をsequenceした。その蛋白のsequenceは今まで知られているものとC末端(V_βとinteractionする場所)が異なった。他のスーパー抗原と比較してV_β3特異的スーパー抗原と最も類似性が高かった。 (3)V_β8.2とV_β8.3の両方に反応するモノクローナル抗体と、V_β8.3のみに反応するものを樹立した。これらの抗体はCzechIIマウスのV_β8.3と反応しないがF23.1抗体とは反応するT cellsの存在により、V_β8.3にはpolymorphismが存在することが判明した。現在、CzechIIマウスよりV_β8.3^+T hybridomaを作成しV_β8.3 sequenceを決定中である。17系統のマウスをこれらの抗体で調べたが、これらのマウスにはV_β8.2,V_β8.3に対するスーパー抗原は存在しなかった。 (4)V_β4に対する他のスーパー抗原、V_β8.2に対する内因性スーパー抗原についてもそのsequenceを決定中である。これらについては既に、Southern blotにより、そのスーパー抗原遺伝子を同定済である。 (5)V_β4,V_β8.2を発現するT hybridoma cellsを作製した。これらV_βに対するスーパー抗原遺伝子のトランスフェクションをして、これらT hybridoma cellsの活性化により特異性の証明を行っている。 (6)本来V_β4特異的スーパー抗原をcodeする遺伝子は内因性のものと考えられた。ところが、この遺伝子が外因性の感染性mouse mammary tumor virusにco-packagingされたことを示す間接的証拠が得られた。さらにRecombinationが感染性mouse mammary tumor virusとの間でおこっているのか否かを検索中である。
