HLA-DR結合性ペプチドモチーフを応用したT細胞エピトープの同定

研究概要	特定のHLA-DR-DQハプロタイプを有するヒトはインスリン依存型糖尿病(IDDM)にかかりやすい。たとえば欧米白人ではDQβ鎖の第57残基(DQβ57)がアスパラギン酸(Asp)であるDQを有する個体はIDDM抵抗性を、また同残基がAspではないDQ遺伝子に関してホモ接合となっている個体はIDDM感受性を示す。そこでDQ分子のペプチド収容溝を構成するα1およびβ1ドメインのアミノ酸配列に関して、DQβ57のみが異なる2つのDQ分子、DQ8(DQβ57;Ala)とDQ(DQβ57;Aps)分子を精製して、両者に結合するペプチドの構造上の特徴の差を解析した。その結果、これらのDQ分子への結合に重要なアンカー残基をposition-1(P-1),P4,P8に同定した。さらにDQβ57と最も近い位置にあるP8アンカー残基のThrを他の19種のアミノ酸に置換して、そのDQ結合性を検討した結果、疎水性アミノ酸は2つのDQ分子に許容されたが、親水性アミノ酸の多くはIDDM感受性を示すDQ8分子にはよく結合したがDQ9分子への結合性は著しく低下した。つまりDQβ57Ala→Aspの多型は、結合ペプチドのC端末側のDQアンカー残基の制約に大きな影響を与えることが明らかとなった。近年白人のIDDM患者より膵臓ランゲルハンス島β細胞に特異的に発現するGAD65に自己反応性を示すT細胞の存在が報告され、その病因との因果関係が注目されている。そこで抗GAD自己抗体陽性の日本人IDDM患者の末梢血リンパ球をGADペプチドと共培養して、多数のGAD自己反応性T細胞クローンを樹立した。今後、GADペプチドを自己反応性T細胞に提示しているHLAクラスII分子を特定し、IDDM感受性とHLAとの関連を解析する予定である。
