乾癬表皮におけるcornified cell envelope形成機構の研究

研究概要

本年度は、主に乾癬表皮のcornified cell envelopeの形成におけるインボルクリンの役割を、免疫電顕を用いて形態面から検討した。その結果、正常表皮ではcornified cell envelopeの形成は角層下方ないし角層直下で起こること、その際、インボルクリンは下方のcornified cell envelopeでは抗原性が保たれているが、角層の中上方では抗原性が消失してしまうことが示された。おそらく正常表皮ではインボルクリンの沈着後、ロリクリン、spr/cornifinといった他のcornified cell envelope構成成分が、順番に沈着していくため、インボルクリンの抗原性のマスキングが起こっているものと推定される。これに合致して、通常電顕におけるcornified cell envelopeの電子密度も、角化の進行とともに変動していくことも示された。以上の結果は、cornified cell envelopeの形成過程が決して瞬間的なものではなく、秩序だった進行性のプロセスであることを示すものである。また、これは同時にcornified cell envelopeの形成が、transglutaminaseの活性化の時点での手持ちの材料によるとする従来のdustbin仮説を否定する根拠にもなっている。一方、乾癬表皮ではcornified cell envelopeの形成は、表皮のより下方から始まり、インボルクリンの抗原性は角層上方まで維持されていた。またcornified cell envelopeの電子密度の変動も認められなかった。したがって、乾癬では、インボルクリン主体のcornified cell envelopeの形成がより未分化な段階の表皮細胞でなされてしまい、その後、他の構成成分による修飾は、あまり起こらないものと考えられた。これは乾癬表皮のcornified cell envelopeがインボルクリン主体の構成要素からなるという従来の推定を支持する所見である。以上の結果は、Journal of Investigative Dermatologyに投稿し、現在、印刷中である。
