表皮内T細胞の機能と活性化機構

研究概要	今年度の研究は主にヒト固定薬珍部表皮に存在するT細胞に関して多くのサンプルを用いて検討を行った。病変部から得られたT細胞に多くはαβTCRを発現しており、CD8^+が多かったがCD4^+も少数認められた。機能についての解析結果は昨年度と同様、NK活性は示さなかったものの、CD3/TCRを介した刺激により様々なtarget cell(keratinocyteを含む)に対し著明な細胞傷害活性を示した。これらのT細胞の抗原特異性は、調べ得た限りにおいて薬剤抗原を認識するものは認められず、自己反応性を示すものが多かった。サイトカイン産生能に関しては、IFN-γ;IL-2,TNF-β産生能をもつT_H1タイプのものが殆どであったが、IL-4,IL-10産生能を示すT_H2タイプのものも確められた。とくに皮疹の新しさや、経過との有意な相関は認められなかった。また、これらの表皮内T細胞はICAM-1を発現した培養ケラチノサイトとの結合のみである程度活性化され、サイトカイン産生に至ることがわかった。 TCRレパートリーに関しても昨年度から引き続き解析を行ったが、やはり著明な偏りが認められとくにVαに関しては皮疹を繰り返したもの程、著明な偏りがあることがわかった。これら固定薬疹に常圧するT細胞は機能的にもheterogeneousであり、それらが微妙なバランスを保ちつつ、表皮のhomeostasisに貢献しているものと考えられる。現在、正常表皮に存在するT細胞に関しても同様の解析を継続中である。
