自己免疫性TSH受容体疾患の概念、成因および自己免疫機構の分析

研究概要	TSH受容体抗体のうち、甲状腺刺激活性を示すTSAbの構造と機能、さらにTSH受容体上の作用部位について研究し、以下の成績を得た。 (1)TSH受容体特異構造部位(アミノ酸no.354-367)に合致するペプチドと、他の受容体ペプチドの混合実験により、受容体上の複数部位が関連してTSAbのエピトープを構成していることを認めた。(森、須川) (2)特異構造部位のペプチドに対するモノクローナル抗体を作製し、この一部がTSAb活性を持つこと、さらに、上述の他受容体部位ペプチドと交叉反応性を示すことを認め、デタ-ミントスプレディング現象と考えた。(須川、森) (3)上記部位を欠失した変異DNAを移植した細胞は、持続的活性化を示した。(小杉、森) 以上の成績から、当該特異構造部位のTSAB産生上の意義が明らかとなった。 (4)患者Bリンパ球をクローニングし、TSAb、TSBb産生クローンを得、この構造解析を行ったが、これによって得られたV_HおよびV_K領域DNAのリコンビネーションを行い、マウスミエローマ細胞を用いて人工的にヒトTSAbを産生する系を確立した。未発表ではあるが、TSAAbのリコンビナント作製にも成功した。(赤水、森) 以上は、TSH受容体の作用、産生機序等について意義深い知見である。
