悪性グリオーマの分子生物学と新しい治療体系への応用

研究概要	すでに私たちはplatelet-derived growth factor(PDGF)に依存性に増殖するグリオーマ細胞に対してPDGF阻害剤であるトラピジ-ルが増殖抑制効果を示すことを明かにした。平成6年度においてはこの増殖因子阻害剤によるグリオーマ細胞内情報伝達系の活性化抑制機構を解明する目的で一連の実験を行なった。この結果、トラピジ-ルにより増殖刺激後のグリオーマ細胞のprotein kinase Cの活性化が抑制され、さらにearly immediate geneのc-fosの発現が抑制されることを明かにした。増殖因子阻害剤の一つであるスラミンは高濃度ではグリオーマ細胞の増殖を抑制したが、低濃度では逆にグリオーマ細胞の増殖を促進し、細胞情報伝達系ではmitogen-activated protein(MAP)kianseの活性を刺激することが明かになった。またトラピジ-ルによりグリオーマ細胞から分泌されるmonocyte chemoattractant protein-1(MCP-1)の産生を抑制する作用があり、グリオーマ局所での免疫反応にも増殖因子阻害因子がなんらかの影響を与えることが明らかになった。同年度に作製予定であったNF-1,NF-2タンパクに対する抗体およびNF1,NF2 geneに対するantisens oligomerに関しては現在進行中である。さらに私たちは、グリオーマ細胞由来のマクロファージ遊走因子(MCP-1)を分離・精製したが、このMCP-1がグリオーマの悪性度と相関し、悪性度が増すにつれMCP-1の発現が強くなることを免疫染色およびノーザンブロット法にて明らかにした。またMCP-1非発現細胞にMCP-1遺伝子を導入することにより、局所へのマクロファージの浸潤が多くなることが明らかになり、今後MCP-1を用いたグリオーマに対する新しい遺伝子治療法の可能性が示された。
