妊娠維持機構における羊膜および脱落膜細胞の役割とその破綻に関する生化学的研究

研究概要

我々は昨年度までの研究で、ヒト羊膜細胞がendothelin(ET)およびbrain natriuretic peptide(BNP)など平滑筋の収縮や弛緩に関与する物質を産生すること、および、様々な細胞で情報伝達に関与していることが知られているphospholipase D(PLD)が羊膜組織にも存在することを報告してきた。本年度は、羊膜、脱落膜および子宮筋におけるETおよびBNPの受容体の発現、およびPLDの生理的意義について胎児成熟との関連から検討し、以下の成績を得た。
1.妊娠末期・中期いずれも絨毛膜、脱落膜、子宮筋には大量のET-AおよびET-B受容体が検出された。妊娠末期の羊膜組織には結合実験でもNorthern blot解析でもET受容体は検出されなかったが、妊娠中期の羊膜組織ではET-A受容体が検出され、培養羊膜細胞ではET-B受容体が検出された。
2.培養羊膜細胞のET-B受容体遺伝子発現は、胎児成熟に伴って羊水中に増加するEGFにより抑制された。
3.絨毛膜、脱落膜、および子宮筋には大量のANP-A受容体が存在し、これらの組織の膜分画にBNPを添加するとsecond messengerであるcGMP産生が増加した。羊膜細胞にはANP-A受容体は発現していなかった。
4.ラット妊娠子宮筋を用いて、in vitroにてPGF2αにより収縮を誘発し、そこにBNPあるいはcGMPを添加すると、いずれも用量依存的に収縮が抑制された。すなわち、ラット妊娠子宮筋には、BNP受容体が存在し、かつcGMPがG-kinaseを介して子宮筋を弛緩していることが示された。
5.羊膜細胞の培養系に、PLDのsecond messengerであるホスファチジン酸(PA)を添加すると、上清中へのPGE2産生量が濃度依存的に増加した。このPGE2産生促進作用はアラキドン(AA)を含まないPAの添加でも同様に見られたことから、PAの中に含まれるAAが基質となるのではなく他の燐脂質からのAAの遊離を促進している可能性が考えられた。脱落膜細胞のPGF2α産生量は有意の増加は見られなかった。
6.PAのPGE2産生促進作用は、protein kinase C(PKC)の阻害剤であるstaurosporineにて抑制されたことから、PKCを介した作用機序が関与していると考えられた。
7.羊膜細胞のPLD活性は、AAやphospholiase C(PLC)にて促進されたことから、羊膜のPLA2,PLC,PLD間にはcross-talkが存在すると考えられた。
